This really brought me down, the repair was expensive. No one else in the country could help me obtain my data and this was definitely my largest #LabFail! Do you have any similar stories? The lab is a very suitable place for gruesome fails!

The gel caught fire on the upper right corner, where the contact from the power source goes in. It was burning under the buffer! The glass broke and having two days of eork left, the only way I could generate my last results was ruined. This set me back 3 months in the project.

It’s #StoryTelling time. This evidently is a very complex system and it requires quite a bit of power (100W+). While running my second ti last gel when suddenly, IT CAUGHT FIRE 🔥 A resistance in the poeer source blew up and couldn’t control the input, creating a short circuit. twitter.com/realscientists…

At least in Central America, we are trying to build our capacities for #ConservationGenetics, from lab skills, bioinformatics, networking and equipment. Nope. The size I’m referring to is the size of these regions of the genome. We measure it in base pairs and ee used the gels to do that, using a DNA “ladder” with several fragments of known sizes, which help us correlate the distance travelled by the fragments with its size.

Our research continues today, trying to find and protect those populations that are not under formal protection and trying to build our knowledge on the genetic diversity of the species as a whole.

What is clear now is that we might have the isolated populations in the best quality of forests, but if we don’t do something to boost genetic diversity, we might lose this important and elusive species from the tree of life.

Now, our plan is to turn and look at the other populations in search of genetic diversity that might revitalize isolated populations. We know that we must reestablish gene flow between populations and aid in the nourishment of this diversity so that populations may adapt.

Captivity individuals and the individual from a distinct population also showed us a light at the end of the tunnel. There were alleles that did not correspond to the most protected population, which meant that other populations might have different alleles.

This species became a candidate for genetic rescue. Not finding a bottleneck event (a population reduction in size and diversity that hinders the population’s adaptability) told us that isolation of the populations was recent, but dangerous.

This meant that the most protected population of this species in Guatemala is struggling with low genetic diversity and might not be able to adapt in the worst scenarios or climate change. This helped me establish urgent investigation needs and a strategy.

And they were what we kind of expected. Genetic diversity was low, an endogamic process was underway, genotypes expressed less heterozygosity than the expected, and we found no evidence of a bottleneck event, curiously.

I did many of these large gels, MANY more smalls gels and failed TONS of PCR runs. It was a constant try-and-fail process since the DNA extraction to the PAGE runs, and after two years of work, many many fails and also exciting field work, WE HAD RESULTS!

The products must be denatured by thermal shock before running and the gel must have a temperature pf 54 degrees C to keep the products denatured. The run is four hours long and the gel must be dyed with silver, which is another compex process all by itself.
