We found that DEXSeq on scaledTPM could find DTU. Two notes are that we did both count and PSI filtering using DRIMSeq functions and this approach is much faster than running on exonic parts
|

I think nearly all the deconvolution method papers talk about this. the log is not distributive, log(a + b) =/= log(a) + log(b). i think it's relevant for whenever you are trying to figure out mixtures. unmix() was written for a collaboration and has no paper associated with it

if you use NNLS for deconvolution, you should be adding expression profiles on the raw expression scale, not log scale. we have a helper function unmix() for making comparisons in log-like scale while doing the NNLS on the raw, easy to copy the code and modify
