    Second big surprise – copy number alterations in these cancer genes are prognostic, even when mutations in those genes are not. For instance, lung cancer patients with/without KRAS mutations have identical outcomes, but KRAS amplifications are associated with patient death.

    We analyzed ~20,000 patients to identify genetic changes associated with patient outcome. First big surprise – most mutations don’t actually affect outcome that much. Mutations in KRAS, PIK3CA, CDKN2A, ATM, SMAD4, etc are commonly observed but rarely prognostic.
